No as the two serial lines are in parallel basically and not crossed over.... TX to TX, RX to RX so one header is used.....

Hi, would there be a possibility of ESP/328P Software Serial communication on io pins ?

I don't see why not.
There is both Serial and SoftwareSerial available on the ESP8266.
